Can my ex stop me from taking a work trip?

Well, sounds like he is being unreasonable, but it is your problem to solve, because you are going on a work trip when you have custody. You will have to find some alternative child care (and pay for it) if he will not cooperate. Your "agreement" as you have described it gives him veto power on your trips. Based on what you posted, I don't think you have any way to force him to accommodate your needs on this trip or any other trip, vacation or otherwise. Do you have a court order or just an "agreement?" If it is just an agreement that has not been incorporated into a court order, it may be in your interest to get a court order on custody and visitation, and make sure you cover all eventualities in it, so that you have an easier way to force him to cooperate in the future. But getting a court order with that kind of complexity is not a DIY project--you will need the help of a lawyer for that.
